چکیده

In this study, we reported the effect of the essential oil of Ocimum gratissimum on Herpetomonas samuelpessoai, a nonpathogenic trypanosomatid. Parasites were grown at 28 or 37°C, in a chemically defined or a complex medium, containing essential oil obtained from Ocimum gratissimum. At concentrations from 20 to 250 μg/ml, the essential oil, progressively inhibited the protozoan growth. The IC50, in defined and complex media, at 28°C were 100 and 91 μg/ml, respectively. Cells cultivated in chemically defined medium were more sensitive to essential oil at concentration of 50, 62.5 and 100 μg/ml in relation to those cultured in complex medium at 37°C. In addition, ultrastructural and enzymatic alterations of the trypanosomatid were also evaluated. H. samuelpessoai exposed to 100 μg/ml of essential oil, in chemically defined medium at 28°C for 72 h, presented considerable ultrastructural alteration, mainly at mitochondrial level, as showed by transmission electron microscopy. Furthermore, cells cultivated in the presence of 100 μg/ml of essential oil showed a decrease of activity of the succinate cytochrome c reductase enzyme, a typical mitochondrion marker, as compared to untreated cells.

Sedative, anxiolytic and antidepressant-like effects of inhalation of the essential oil of Ocimum gratissimum L. from Cameroon in mice

In this study, the behavioral effects of inhalation of the essential oil of Ocimum gratissimum from Cameroon were investigated. The open field test, light/dark box test, tail suspension test and Rota-rod test were used to assess the sedative, anxiolytic-like, antidepressant-like and motor coordination effects respectively, in mice. GC and GC/MS analyses were performed to investigate the chemica...

Effect of sphingosine and phorbol-12-myristate-13-acetate on the growth and dimethylsulfoxide-induced differentiation in the insect trypanosomatid Herpetomonas samuelpessoai.

We investigated the effect of two modulators of protein kinase C, sphingosine and phorbol-12-myristate-13-acetate (PMA), on the growth and dimethylsulfoxide (DMSO)-induced differentiation in Herpetomonas samuelpessoai. Sphingosine did not stimulate the transformation of undifferentiated-promastigotes in differentiated-paramastigotes. PMA alone or in association with DMSO increased the number of...

Isolation of a protozoan parasite genetically related to the insect trypanosomatid Herpetomonas samuelpessoai from a human immunodeficiency virus-positive patient.

Severely immunocompromised human immunodeficiency virus (HIV) patients can develop various opportunistic infections due to bacteria, viruses, fungi, or protozoa. Here we report the first isolation of a flagellated protozoan genetically closely related to Herpetomonas samuelpessoai, which is usually a parasite of insects, from the blood of an HIV-infected patient.

Chemical Composition, In Vitro Antimicrobial and Antioxidant Activities of the Essential Oils of Ocimum Gratissimum, O. Sanctum and their Major Constituents

The essential oils of the flowering aerial parts of two Ocimum species viz., Ocimum gratissimum and O. sanctum were analyzed by gas chromatography and gas chromatography/mass spectroscopy. The principal constituent of O. gratissimum and O. sanctum was eugenol (75.1%) and methyl eugenol (92.4%), comprising 99.3 and 98.9% of the total oils, respectively. In vitro antimicrobial activity of the ess...
